產(chǎn)品簡介:

植物原生質(zhì)體是指脫去全部細(xì)胞壁由質(zhì)膜包被的具有生命活力的裸露細(xì)胞。它具有細(xì)胞生命特征和全能型,是細(xì)胞無性系變異和突變體篩選的重要來源,同時也是植物遺傳工程的理想受體和遺傳改良的理想材料。酶解法分離原生質(zhì)體是一個常用的技術(shù),其原理是植物細(xì)胞壁主要由纖維素、半纖維素和果膠質(zhì)組成,因而使用纖維素酶、半纖維素酶、離析酶和果膠酶能降解細(xì)胞壁成分,去除細(xì)胞壁,即可得到原生質(zhì)體。許多方法可誘導(dǎo)原生質(zhì)體融合,現(xiàn)在被廣泛采用的融合方法是聚乙二醇(PEG)法。聚乙二醇(PEG)作為一種高分子化合物,合適的濃度能對原生質(zhì)體產(chǎn)生瞬間沖擊效應(yīng),原生質(zhì)體很快發(fā)生收縮與粘連,隨后用高鈣高pH法進(jìn)行清洗,使原生質(zhì)體融合得以完成。

按照每次使用5 ml酶消化體系計算,本試劑盒可用于40次酶消化反應(yīng)。本試劑盒每個5 ml反應(yīng)體系可處理0.1 g左右的擬南芥葉片(約10~15葉片),操作較好的情況下每5 ml體系可獲得約50-70萬個原生質(zhì)體(不同植物不同操作會有一定的差異),可滿足約25-70個樣品的原生質(zhì)體質(zhì)粒轉(zhuǎn)染操作(按照每個樣品1-2萬個細(xì)胞計算)。

原生質(zhì)體轉(zhuǎn)化時,本試劑盒可以用于相當(dāng)于6孔板40個孔的樣品,12孔板80個孔的樣品,或24孔板160個孔的樣品的轉(zhuǎn)化。

貯存和效期:

-20°C保存,至少一年有效。

組分III,III,V  4℃存放3-5天不會影響使用效果,長期不用,按照標(biāo)簽-20℃貯存。

試劑盒常溫運(yùn)輸。

使用說明:

   需要準(zhǔn)備的材料(試劑盒不提供):

   剪尖吸頭(剪尖后可以用酒精燈過火將吸頭處理平滑);平頭鑷子;70 μm細(xì)胞過濾篩;一次性刀片;50 ml離心管;1.5 ml離心管;水浴鍋。

注:即用型酶溶液現(xiàn)用現(xiàn)配,不建議配制后凍存后再使用;

溶解好的正常酶溶液應(yīng)為澄清棕黃色溶液,如使用純度不好的酶,溶液為不溶解的乳白色懸濁液,不能使用。

1. 酶解:

取生長狀態(tài)良好的葉片切成0.5-1 mm的小條,按照0.1 克葉片(擬南芥約15-20個葉片)加5 ml即用型酶溶液的比例迅速將切割好的葉片小條浸泡于酶溶液中,避光,無需震蕩,常溫(20-25℃)酶解3小時,間歇混勻。

注:酶解時間與葉片種類,葉片生長狀態(tài)有關(guān),請根據(jù)實驗需要適當(dāng)調(diào)整酶解時間。3小時為擬南芥葉片推薦的酶解時間。如條件允許,可以使用微型真空泵常溫避光條件下抽真空30 min,以使酶溶液更好地進(jìn)入細(xì)胞間隙。酶溶液變?yōu)榫G色表明有原生質(zhì)體已經(jīng)有分離,溶液為濃綠色表明原生質(zhì)體已經(jīng)大量分離。擬南芥原生質(zhì)體大小約為30-50 μm,顯微鏡鏡檢后確定是否分離。葉片原生質(zhì)體細(xì)胞顯微鏡下為綠色圓球狀,葉綠體分散在整個細(xì)胞內(nèi),說明狀態(tài)較好;如呈現(xiàn)不規(guī)則形狀,說明原生質(zhì)

體破碎或即將破碎。


 

 

  2. 漂洗:

酶解后加入等體積的溶液II,如使用5 ml酶解體系,加入5 ml溶液II,輕柔混勻。

3. 過濾:

用孔徑70 μm篩網(wǎng)過濾步驟2中的溶液,去除未消化的葉片,收集濾出液于50 ml離心管中。

4. 第一次收集:

濾出液100g常溫離心 2分鐘,盡量去除上清。

注:為了避免原生質(zhì)體離心時貼在管壁,建議整個實驗過程使用水平轉(zhuǎn)頭;離心時,可調(diào)低離心機(jī)的升速和降速。升速過快,原生質(zhì)體可能離到管壁上;降速過快,可能導(dǎo)致管底原生質(zhì)體懸起。

5. 第二次收集:

加入2-5 ml 溶液II,用剪尖的藍(lán)吸頭重懸原生質(zhì)體,冰浴30分鐘,原生質(zhì)體在重力作用下可以沉降到離心管底部,盡量吸除上清,收集原生質(zhì)體。(如果發(fā)現(xiàn)原生質(zhì)體沉降的速度比較慢或者得率比較低,也可以考慮常溫100 g離心1-2 min收集原生質(zhì)體)。

6. 重懸:

小心去除上清溶液,不要觸動原生質(zhì)體沉淀,沉淀用剪尖的藍(lán)吸頭重懸于1 ml溶液III中即為原生質(zhì)體溶液。(可以用血球計數(shù)板計數(shù),根據(jù)原生質(zhì)體數(shù)量調(diào)整溶液III的加入體積,使得原生質(zhì)體密度為2×105/ml或更高)。

注:制備好的原生質(zhì)體可以在4°C或冰浴保存至少24 h。

二、原生質(zhì)體轉(zhuǎn)化和培養(yǎng):

  1. 轉(zhuǎn)化溶液配制:

植物原生質(zhì)體轉(zhuǎn)化參考下表根據(jù)樣品量配制轉(zhuǎn)化溶液。

轉(zhuǎn)化溶液現(xiàn)用現(xiàn)配。轉(zhuǎn)化溶液需要在轉(zhuǎn)化前至少1小時配制,以確保轉(zhuǎn)化試劑溶解充分。轉(zhuǎn)化溶液配制后盡量當(dāng)天使用。配制好的轉(zhuǎn)化溶液4℃保存3-5天之內(nèi)仍然有較好的轉(zhuǎn)化效率,但和當(dāng)天配制的轉(zhuǎn)化溶液相比,轉(zhuǎn)化效果可能會有一定程度的下降。

2. 準(zhǔn)備質(zhì)粒:

取10 μl質(zhì)粒DNA(10-20 μg,濃度為1-2 μg/μl)于1.5 ml 離心管中。

注:質(zhì)粒大小建議為5-10 kb,質(zhì)粒濃度為1-2 μg/μl左右;

原生質(zhì)體轉(zhuǎn)化對于質(zhì)粒的純度要求較高,盡量使用高純度的質(zhì)粒。

3. 質(zhì)粒轉(zhuǎn)化:

3.1 質(zhì)粒管中加入100 μl原生質(zhì)體溶液(原生質(zhì)體密度為2×105/ml,約2萬個原生質(zhì)體),輕柔混勻。

3.2 加入等體積即110 μl 步驟1事先準(zhǔn)備好的轉(zhuǎn)化溶液,輕彈管底,輕柔混勻,常溫25℃放置5-15分鐘。

注:最長可以孵育15 min,但通常孵育5min時間已經(jīng)足夠。最佳的孵育時間對于不同的原生質(zhì)體和不同的質(zhì)粒需要通過實驗摸索。

4. 終止轉(zhuǎn)化:

加入2倍體積即440 μl 溶液II,輕柔徹底混勻,終止轉(zhuǎn)化過程。

5. 收集原生質(zhì)體:

常溫100g離心1-2分鐘,盡量去除上清。

注:由于轉(zhuǎn)化后的溶液會非常粘稠,加入溶液II后離心1 min通??梢允乖|(zhì)體聚集在管底,但使用某些突變體時為減少原生質(zhì)體損失,可將離心時間延長到2 min。

6. 原生質(zhì)體漂洗:

  加入500 μl溶液II用剪尖藍(lán)吸頭輕輕重懸原生質(zhì)體,常溫100g離心1 min,盡量去除殘留的上清。

注:本步驟可以充分去除殘留的轉(zhuǎn)染試劑溶液中的組分,避免轉(zhuǎn)染試劑溶液中的組分對于后續(xù)的不良影響。

7. 原生質(zhì)體重懸:

沉淀中加入1 ml溶液V,用剪尖藍(lán)吸頭輕柔重懸。

8. 原生質(zhì)體培養(yǎng):

將離心管水平放置,23-25°C弱光培養(yǎng)。

注:根據(jù)實驗需求確定孵育時間。RNA分析孵育2-6小時;酶活性分析和蛋白標(biāo)記實驗孵育2-16小時;基因編輯的效果在轉(zhuǎn)染24小時后可能被檢測到。

三、實驗示例:

 

使用原生質(zhì)體植物原生質(zhì)體制備及轉(zhuǎn)化試劑盒轉(zhuǎn)染擬南芥原生質(zhì)體的效果圖。

實驗步驟:稱取0.48 g轉(zhuǎn)化試劑于2 ml 離心管中,加入轉(zhuǎn)化試劑溶解液后,顛倒混勻,蒸餾水定容至2 ml,使轉(zhuǎn)化試劑充分溶解后備用。在2 ml的圓底離心管中加入10 μl(20 μg)EGFP質(zhì)粒 (植物用綠色熒光蛋白),加入100 μl制備好的原生質(zhì)體,輕柔混勻后加入110 μl當(dāng)日配制好的轉(zhuǎn)化試劑溶液,輕柔混勻,常溫靜置5 min后加入440 μl溶液II終止轉(zhuǎn)化,輕輕顛倒混勻,常溫100 g離心1 min,去除上清,再加入0.5 ml溶液II,輕柔重懸原生質(zhì)體,常溫100 g離心1 min后,盡量去除上清,收集原生質(zhì)體。加入1 ml溶液V,小心重懸原生質(zhì)體后水平放置25℃培養(yǎng)過夜(約16h),次日于熒光顯微鏡下檢測EGFP熒光信號。
